Department/Project:

The Vaccines department is working on the discovery- and early development of novel vaccines for bacterial infectious diseases. We strive to address unmet medical needs, such as bacteremia (sepsis) and nosocomial infections. Projects at the Bacterial Vaccines department are primarily structured on a scientific- and technical collaboration with external biotechnology partners (governmental and industrial) that are innovative leaders in their field. This working process includes ‘state of the art’ vaccine discovery, with subsequent in vivo evaluation of these vaccine candidates in animal models for immunogenicity and/or efficacy. In addition, manufacturing processes are optimized and established, and the resulting clinical trial material is evaluated in human clinical studies.

 

Job Description:

You will be part of a dynamic team driving research-scale production and characterization of bacterial vaccine candidates, mainly consisting of protein- and glycoprotein antigens. 

To this end, the  Downstream Process Development team develops and characterizes a broad range of biopharmaceutical DSP units using, amongst others, high-throughput process development methodologies and develops end-to-end purification processes at different scales. 

These newly developed purification processes are used for producing research-grade material for preclinical evaluation and are transferred to early process development partners.

As an Assistant Scientist DSP, you will support the Downstream Process Development team in performing purification experiments and in-process monitoring assays to facilitate early-stage bacterial vaccine development.

 

Your main responsibilities will be:

  • Supporting and executing purification and filtration experiments.
  • Preparation and execution of (high throughput) in-process monitoring assays.
  • Transferring in-process monitoring assays (plate-based) to high-throughput platforms.
  • Performing (preliminary) data analysis, reporting, and presentation of the main conclusions to the DSP team.
  • Experimental documentation in an electronic laboratory notebook (ELN).
  • Preparation of process documentation, including work instructions and batch records.
  • Handling laboratory stocks and keeping inventories up to date.
  • Keeping track of equipment status and/or maintenance.
  • Keeping systems in place for inventory/tracking of samples and process intermediates.
  • Following safety procedures, contribute to the maintenance of a clean laboratory environment.

 

Education:

  • Bachelor or Master degree or equivalent, in a relevant field such as Biochemistry, Bacteriology, Biomedical Sceinces, Biotechnology, Biological Engineering or related fields.

 

Experience:

  • 0-2 years relevant experience or relevant internships.
  • Experience with protein and/or glycoprotein purification (DSP) methodologies.
  • Understanding of analytical techniques to analyze proteins (e.g. plate-based colorimetric assays, capillary electrophoresis, ELISA, Western blotting, BLI, SDS-PAGE, UPLC-based techniques).
  • Knowledge of high-throughput platforms and liquid handling systems.

 

Competences:

  • High affinity for lab work.
  • Affinity with automation of processes and assays for high-throughput applications.
  • Theoretical understanding of purification technologies (e.g. chromatography, filtration, precipitation) and/or analytical techniques to analyze large molecules (e.g. proteins, polysaccharides, glycoproteins).
  • Flexible and creative team player.
  • Communicates clearly and efficiently.
  • Is organized and methodic in data generation and handling (e.g. security and integrity).
  • Language skills: Fluent in English (and Dutch).
  • Relevant software skills (e.g. Unicorn, MS Office, databases, sample management).

 

EXTRA INFORMATION:

  • You will work on-site in Leiden.
  • This is a contracting position, with the possibility of an extension after the end date. You will receive a contract via our partner YACHT. They can offer you a contract for the duration of the assignment at J&J, a one year contract or a freelance contract (ZZP).
